Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_170675.5(MEIS2):c.640-29G>A, citing Ambry Variant Classification Scheme 2023: The c.640-29G>A intronic alteration results from a G to A substitution 29 nucleotides before coding exon 7 of the MEIS2 gene. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
