Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004991.4(MECOM):c.2600C>T (p.Thr867Ile), citing Ambry Variant Classification Scheme 2023: The p.T867I variant (also known as c.2600C>T), located in coding exon 10 of the MECOM gene, results from a C to T substitution at nucleotide position 2600. The threonine at codon 867 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
