Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006767.4(LZTR1):c.1936G>T (p.Asp646Tyr), citing Ambry Variant Classification Scheme 2023: The p.D646Y variant (also known as c.1936G>T), located in coding exon 16 of the LZTR1 gene, results from a G to T substitution at nucleotide position 1936. The aspartic acid at codon 646 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
