This variant lies in the ABCA12 gene (transcript NM_173076.3) at coding-DNA position 2968, where A is replaced by G; at the protein level this means replaces lysine at residue 990 with glutamic acid — a missense variant. Submitter rationale: The K990E variant in the ABCA12 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. K990E was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. It is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals. However, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret K990E as a variant of uncertain significance.
