This variant lies in the CACNA1H gene (transcript NM_021098.3) at coding-DNA position 2146, where G is replaced by T; at the protein level this means replaces glycine at residue 716 with cysteine — a missense variant. Submitter rationale: The G716C variant in the CACNA1H gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G716C variant was not observed in approximately 6400 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The G716C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret G716C as a variant of uncertain significance.
