Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_031407.7(HUWE1):c.7204+1G>T, citing Ambry Variant Classification Scheme 2023: The c.7204+1G>T intronic variant results from a G to T substitution one nucleotide after exon 53 (coding exon 50) of the HUWE1 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, loss of function of HUWE1 has not been established as a mechanism of disease. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
