NM_020987.5(ANK3):c.6320C>G (p.Thr2107Ser) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The T2107S variant in the ANK3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The T2107S variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret T2107S as a variant of uncertain significance.
