Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_013372.7(GREM1):c.238G>C (p.Ala80Pro), citing Ambry Variant Classification Scheme 2023: The p.A80P variant (also known as c.238G>C), located in coding exon 1 of the GREM1 gene, results from a G to C substitution at nucleotide position 238. The alanine at codon 80 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
