NM_013372.7(GREM1):c.313G>C (p.Glu105Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E105Q variant (also known as c.313G>C), located in coding exon 1 of the GREM1 gene, results from a G to C substitution at nucleotide position 313. The glutamic acid at codon 105 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
