NM_004304.5(ALK):c.3755C>A (p.Ala1252Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A1252D variant (also known as c.3755C>A), located in coding exon 25 of the ALK gene, results from a C to A substitution at nucleotide position 3755. The alanine at codon 1252 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
