Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.698G>A (p.Ser233Asn), citing Ambry Variant Classification Scheme 2023: The p.S233N variant (also known as c.698G>A), located in coding exon 5 of the GEN1 gene, results from a G to A substitution at nucleotide position 698. The serine at codon 233 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
