Uncertain significance — the classification assigned by Ambry Genetics to NM_024642.5(GALNT12):c.413C>G (p.Thr138Arg), citing Ambry Variant Classification Scheme 2023: The p.T138R variant (also known as c.413C>G), located in coding exon 2 of the GALNT12 gene, results from a C to G substitution at nucleotide position 413. The threonine at codon 138 is replaced by arginine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
