This variant lies in the ANK3 gene (transcript NM_020987.5) at coding-DNA position 3575, where T is replaced by A; at the protein level this means replaces isoleucine at residue 1192 with asparagine — a missense variant. Submitter rationale: The I1192N variant in the ANK3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The I1192N variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Isoleucine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret I1192N as a variant of uncertain significance.
