Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.574A>G (p.Asn192Asp), citing Ambry Variant Classification Scheme 2023: The p.N192D variant (also known as c.574A>G), located in coding exon 6 of the FBN1 gene, results from an A to G substitution at nucleotide position 574. The asparagine at codon 192 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
