Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.4112G>C (p.Gly1371Ala), citing Ambry Variant Classification Scheme 2023: The p.G1371A variant (also known as c.4112G>C), located in coding exon 41 of the FANCA gene, results from a G to C substitution at nucleotide position 4112. The glycine at codon 1371 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
