Uncertain significance — the classification assigned by Ambry Genetics to NM_022051.3(EGLN1):c.602G>A (p.Cys201Tyr), citing Ambry Variant Classification Scheme 2023: The p.C201Y variant (also known as c.602G>A), located in coding exon 1 of the EGLN1 gene, results from a G to A substitution at nucleotide position 602. The cysteine at codon 201 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
