Uncertain significance — the classification assigned by Ambry Genetics to NM_022051.3(EGLN1):c.812T>G (p.Leu271Arg), citing Ambry Variant Classification Scheme 2023: The p.L271R variant (also known as c.812T>G), located in coding exon 1 of the EGLN1 gene, results from a T to G substitution at nucleotide position 812. The leucine at codon 271 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
