Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.41T>C (p.Leu14Pro), citing Ambry Variant Classification Scheme 2023: The p.L14P variant (also known as c.41T>C), located in coding exon 1 of the EGFR gene, results from a T to C substitution at nucleotide position 41. The leucine at codon 14 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
