NM_018972.4(GDAP1):c.347T>G (p.Met116Arg) was classified as Pathogenic for Charcot-Marie-Tooth disease; Charcot-Marie-Tooth disease type 4A by Sadaf Naz Human Genetics Laboratory, University of the Punjab, citing ACMG Guidelines, 2015: The variant Met116Arg caused Charcot-Marie-Tooth disease in two individuals of a consanguineous family. The patients cannot walk and move by dragging themselves with hands. They presented amyotrophy and areflexia of distal limb muscles. They exhibit valgus wrists and ankles and foot contractures. The variant c.347T>G, p.Met116Arg had an extremely low frequency in gnomAD and was absent in the 600 chromosomes of ethnically matched control population. The variant was previously reported to cause recessively inherited CMT in four Italian individuals in three different families. In summary, we classify the variant as pathogenic based on our segregation analysis and absence of the variant in ethnically matched control population.
