Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.8949G>C (p.Leu2983Phe), citing Ambry Variant Classification Scheme 2023: The p.L2983F variant (also known as c.8949G>C), located in coding exon 55 of the DNAH11 gene, results from a G to C substitution at nucleotide position 8949. The leucine at codon 2983 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
