Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.2249C>A (p.Pro750Gln), citing Ambry Variant Classification Scheme 2023: The p.P750Q variant (also known as c.2249C>A), located in coding exon 13 of the DICER1 gene, results from a C to A substitution at nucleotide position 2249. The proline at codon 750 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
