This variant lies in the ANK3 gene (transcript NM_020987.5) at coding-DNA position 7925, where C is replaced by G; at the protein level this means replaces serine at residue 2642 with cysteine — a missense variant. Submitter rationale: The S2642C variant in the ANK3 gene has not been reported previously as a pathogenic variant, nor as a benignvariant, to our knowledge. The S2642C variant was not observed in approximately 6500 individuals of European andAfrican American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant inthese populations. The S2642C variant is a non-conservative amino acid substitution, which is likely to impactsecondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitutionoccurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not thevariant is damaging to the protein structure/function. We interpret S2642C as a variant of uncertain significance.
