NM_001903.5(CTNNA1):c.1052A>T (p.Tyr351Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y351F variant (also known as c.1052A>T), located in coding exon 6 of the CTNNA1 gene, results from an A to T substitution at nucleotide position 1052. The tyrosine at codon 351 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
