Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.417C>A (p.Asn139Lys), citing Ambry Variant Classification Scheme 2023: The p.N139K variant (also known as c.417C>A), located in coding exon 4 of the CPA1 gene, results from a C to A substitution at nucleotide position 417. The asparagine at codon 139 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
