Likely benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002641.4(PIGA):c.-63+8C>T, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PIGA c.-63+8C>T is located in the untranslated mRNA region upstream of the initiation codon. The variant allele was found at a frequency of 5.3e-05 in 982176 control chromosomes, including 19 hemizygotes. This frequency is not significantly higher than estimated for disease-causing variants in PIGA, however the observation of several hemizygous controls is not consistent with the early onset/severe presentation of PIGA-related conditions. To our knowledge, no occurrence of c.-63+8C>T in individuals affected with PIGA-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 383325). Based on the evidence outlined above, the variant was classified as likely benign.
