Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.2909A>G (p.Asn970Ser), citing Ambry Variant Classification Scheme 2023: The p.N970S variant (also known as c.2909A>G), located in coding exon 10 of the CDK12 gene, results from an A to G substitution at nucleotide position 2909. The asparagine at codon 970 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
