Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.3917C>G (p.Ser1306Cys), citing Ambry Variant Classification Scheme 2023: The p.S1306C variant (also known as c.3917C>G), located in coding exon 14 of the CDK12 gene, results from a C to G substitution at nucleotide position 3917. The serine at codon 1306 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
