NM_001232.4(CASQ2):c.83A>C (p.Tyr28Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y28S variant (also known as c.83A>C), located in coding exon 1 of the CASQ2 gene, results from an A to C substitution at nucleotide position 83. The tyrosine at codon 28 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
