Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.2715_2720del (p.Ile906_Leu907del), citing Ambry Variant Classification Scheme 2023. This variant lies in the CACNA1C gene (transcript NM_000719.7) at coding-DNA position 2715 through coding-DNA position 2720, deleting 6 bases. Submitter rationale: The c.2715_2720delGATCCT (p.I906_L907del) alteration, located in coding exon 20 of the CACNA1C gene, results from an in-frame deletion of 6 nucleotides at positions c.2715 to c.2720. This results in the deletion of 2 amino acids between codons 906 and 907. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). These amino acid positions are highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis (Choi, 2012). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
