Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.95C>A (p.Ala32Asp), citing Ambry Variant Classification Scheme 2023: The p.A32D variant (also known as c.95C>A), located in coding exon 2 of the CACNA1C gene, results from a C to A substitution at nucleotide position 95. The alanine at codon 32 is replaced by aspartic acid, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
