This variant lies in the RYR1 gene (transcript NM_000540.3) at 5 bases into the intron immediately after coding-DNA position 3381, where G is replaced by A. Submitter rationale: This variant causes a G to A nucleotide substitution at the +5 position of intron 25 of the RYR1 gene. Splice site prediction tools are inconclusive regarding the impact of this variant on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with autosomal dominant malignant hyperthermia in the literature, although it is associated with other phenotype(s) (ClinVar Variation ID: 382493). This variant has been identified in 2/249156 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Due to insufficient evidence, this variant is classified as a Variant of Uncertain Significance for autosomal dominant malignant hyperthermia.
