Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004329.3(BMPR1A):c.184T>C (p.Tyr62His), citing Ambry Variant Classification Scheme 2023: The p.Y62H variant (also known as c.184T>C), located in coding exon 2 of the BMPR1A gene, results from a T to C substitution at nucleotide position 184. The tyrosine at codon 62 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
