Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.2014G>T (p.Asp672Tyr), citing Ambry Variant Classification Scheme 2023: The p.D672Y variant (also known as c.2014G>T), located in coding exon 16 of the BAP1 gene, results from a G to T substitution at nucleotide position 2014. The aspartic acid at codon 672 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
