Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_024598.4(USB1):c.97C>G (p.Arg33Gly), citing Ambry Variant Classification Scheme 2023: The p.R33G variant (also known as c.97C>G), located in coding exon 1 of the USB1 gene, results from a C to G substitution at nucleotide position 97. The arginine at codon 33 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
