Pathogenic — the classification assigned by Ambry Genetics to NM_019059.5(TOMM7):c.26_29del (p.Lys9fs), citing Ambry Variant Classification Scheme 2023: The c.26_29delAGCA (p.K9Rfs*22) alteration, located in exon 1 (coding exon 1) of the TOMM7 gene, consists of a deletion of 4 nucleotides from position 26 to 29, causing a translational frameshift with a predicted alternate stop codon after 22 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
