Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001365276.2(TNXB):c.797G>A (p.Cys266Tyr), citing Ambry Variant Classification Scheme 2023: The p.C266Y variant (also known as c.797G>A), located in coding exon 2 of the TNXB gene, results from a G to A substitution at nucleotide position 797. The cysteine at codon 266 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
