NM_017849.4(TMEM127):c.353C>G (p.Pro118Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P118R variant (also known as c.353C>G), located in coding exon 2 of the TMEM127 gene, results from a C to G substitution at nucleotide position 353. The proline at codon 118 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
