Uncertain significance — the classification assigned by Ambry Genetics to NM_001127208.3(TET2):c.1018A>G (p.Ile340Val), citing Ambry Variant Classification Scheme 2023: The p.I340V variant (also known as c.1018A>G), located in coding exon 1 of the TET2 gene, results from an A to G substitution at nucleotide position 1018. The isoleucine at codon 340 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
