Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.1976A>C (p.Lys659Thr), citing Ambry Variant Classification Scheme 2023: The p.K659T variant (also known as c.1976A>C), located in coding exon 5 of the TERT gene, results from an A to C substitution at nucleotide position 1976. The lysine at codon 659 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
