NM_000051.4(ATM):c.6101G>T (p.Arg2034Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R2034L variant (also known as c.6101G>T), located in coding exon 41 of the ATM gene, results from a G to T substitution at nucleotide position 6101. The arginine at codon 2034 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
