Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_006940.6(SOX5):c.1093C>A (p.Leu365Ile), citing Ambry Variant Classification Scheme 2023: The c.1093C>A (p.L365I) alteration is located in exon 9 (coding exon 9) of the SOX5 gene. This alteration results from a C to A substitution at nucleotide position 1093, causing the leucine (L) at amino acid position 365 to be replaced by an isoleucine (I). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
