Uncertain Significance for Arrhythmogenic right ventricular cardiomyopathy — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001005242.3(PKP2):c.2013+1G>C, citing ACMG Guidelines, 2015. This variant lies in the PKP2 gene (transcript NM_001005242.3) at the canonical splice donor site of the intron immediately after coding-DNA position 2013, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes a G to C nucleotide substitution at the +1 position of intron 10 of the PKP2 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with arrhythmogenic right ventricular cardiomyopathy (PMID: 28588093). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
