Pathogenic — the classification assigned by GeneDx to NM_001368894.2(PAX6):c.1226-2A>G, citing GeneDx Variant Classification (06012015). This variant lies in the PAX6 gene (transcript NM_001368894.2) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1226, where A is replaced by G; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1184-2 A>G splice site variant in the PAX6 gene destroys the canonical splice acceptor site in intron 12. It is predicted to cause abnormal gene splicing, either leading to an abnormal message that is subject to nonsense-mediated mRNA decay, or to an abnormal protein product if the message is used for protein translation. Although this variant has not been previously reported to our knowledge, we interpret it as pathogenic.
