Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_017654.4(SAMD9):c.2204T>C (p.Leu735Pro), citing Ambry Variant Classification Scheme 2023: The p.L735P variant (also known as c.2204T>C), located in coding exon 1 of the SAMD9 gene, results from a T to C substitution at nucleotide position 2204. The leucine at codon 735 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
