NM_001754.5(RUNX1):c.755C>A (p.Ser252Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S252Y variant (also known as c.755C>A), located in coding exon 6 of the RUNX1 gene, results from a C to A substitution at nucleotide position 755. The serine at codon 252 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
