Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001283009.2(RTEL1):c.416G>A (p.Gly139Asp), citing Ambry Variant Classification Scheme 2023: The p.G139D variant (also known as c.416G>A), located in coding exon 4 of the RTEL1 gene, results from a G to A substitution at nucleotide position 416. The glycine at codon 139 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
