NM_001283009.2(RTEL1):c.2457G>C (p.Glu819Asp) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E819D variant (also known as c.2457G>C), located in coding exon 26 of the RTEL1 gene, results from a G to C substitution at nucleotide position 2457. The glutamic acid at codon 819 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
