Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.2140C>T (p.Pro714Ser), citing Ambry Variant Classification Scheme 2023: The p.P714S variant (also known as c.2140C>T), located in coding exon 14 of the RINT1 gene, results from a C to T substitution at nucleotide position 2140. The proline at codon 714 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
