Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000321.3(RB1):c.1607T>C (p.Ile536Thr), citing Ambry Variant Classification Scheme 2023: The p.I536T variant (also known as c.1607T>C), located in coding exon 17 of the RB1 gene, results from a T to C substitution at nucleotide position 1607. The isoleucine at codon 536 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
