Uncertain significance — the classification assigned by Ambry Genetics to NM_133510.4(RAD51B):c.1043A>G (p.Glu348Gly), citing Ambry Variant Classification Scheme 2023: The p.E348G variant (also known as c.1043A>G), located in coding exon 10 of the RAD51B gene, results from an A to G substitution at nucleotide position 1043. The glutamic acid at codon 348 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
